蛋白質の発現レベルの変化


 老化した動物の細胞や培養条件下で老化した細胞では、細胞内および細胞膜表面の蛋白質の発現レベルが変化している事が観察されています。
 このような『加齢に伴う蛋白質発現レベルの変化』は、『細胞における老化現象』の一つであり、この上流には『老化の根本原因』が潜んでいるものと思われます。またこの下流においては、細胞機能の低下に伴い臓器機能が不全を起こすリスクが高まってくることから、これらの現象は個体の老化を考える上でも極めて重要であると考えられます。
 蛋白質の発現レベルが変化する原因としては、(1)塩基置換やDNAメチル化などの遺伝子上の変化、(2)ヒストンや転写因子などの蛋白質の変化、(3)細胞外マトリクスなどの細胞周囲の微小環境の変化などが考えられますが、それらの変化の背景には酸素ラジカルなどによる酸化的なストレスの影響があるものと考えられます。
